Identifying, cloning and sequencing of a RAPD marker linked to mildew-resistance gene of Hevea brasiliensis

By: Contributor(s): Material type: TextTextPublication details: Paper presented at IRRDB, China, 1999Subject(s): Summary: The genomic DNA isolated from 11 mildew (Oidium hevea)-resistant and 11 susceptible clones of Hevea brasiliensis were analyzed with 52 random primers by using RAPD technique. A DNA fragment designated OPV-10 390 amplified from the genomic DNA from RRTC52 (CK) demonstrated that this sequence was low copy sequence in the resistant clones but not contained in the susceptible clones. It can thus be seen that OPV-10 390 can be used as RAPD markers linked to mildew-resistance gene. OPV-10 390 was cloned and sequenced, and analyzed by Blastn. Results showed that this fragment was a new sequence.
