An Efficient method for the isolation of good quality RNA from bark tissues of mature rubber trees

The method explained here involes removal of phenolic compounds using polyvinyl pyrrolidone and selective precipitation of RNA using lithium chloride. The purity of RNA prepared by this method was tested for its ability to get a good quality cDNA prepared. The cDNA produced from total RNA preparation was further con firmed by amplification of specific house keeping genes using quantitative real time PCR method. This protocol appears best suited for Hevea bark that is rich in several contaminating factors.
