520 _aTwo allergenic (IgE-binding) proteins have been isolated from the B-serum of natural rubber latex by dialysis-induced protein precipitation followed by gel filtration. One of the allergens, Hev b II, that is specific to the monoclonal antibody USM/RB4, commonly appears as a doublet of molecular weights 34 and 36 kDa under reducing conditions of SDS-polycacrylamide gel electrophoresis. In the absence of 2-mercaptoethanol, a 70 kDa band can also be seen in addition to the doublet. The allergen has a pI of circa 9.5 and is identified as B-1, 3-glucanase. The second allergen, Hev b IV, has a pI at of crica 4.5 and appears under under reducing conditions as a band of 50-57 kDa. In its unreduced form, a principal band of approximately 100 kDa is discernible. Immuno-gold labelling with the monoclonal antibody USM/RB3 shows that Hev B IV is a component of the microhelix protein complex of B-serum.
