Summary, etc The presence of numerous bacteria in the phyllosphere of terrestrial plants has been demonstrated, however no reports associated with rubber plant. Here we report a genetic diversity of phyllosphere bacteria and the bacteria, which is usually found, on rubber leaves. In the future the suitable bacteria will be used as an antagonist organism or an engineered-organism against Corynespora leaf fall disease. The bacterial colonies were isolated from two experimental garden i.e. Ciomas (Biotechnology Research Unit for Estate Crops), Bogor and Sembawa (Rubber Research Station), Palembang from upper and lower surfaces of leaf of RRIC 100, PR 255, RRIC 103, and PPN 2444 clones. The first two clones are well known resistant to Corynespora leaf fall disease while the rest are susceptible. Total bacterial DNA was isolated from each colony and the 16S-rRNA was amplified using specific primer of bacterial domain, i.e. 63f and 1387r. Restriction analyses of 16S-rNA genes were done by digesting these fragments with two kinds of tetrameric endonuclease enzyme, i.e. RsaI and HhaI. The result showed that 39 and 37 isolates of the bacteria, from Ciomas and Sembawa respectively, both from upper and lower part of the leaves were successfully isolated from these clones. Some of bacterial isolates have a similar morphology and some were different. Amplification of 16S rRNA genes from all isolates yielded single band with +_ 1300 bp in size. These PCR-products were cleaved by RsaI and HhaI and showed that all isolates had more than one restriction sites. The grouping of rubber phyllosphere bacteria and genetic diversity of these bacteria based on the PCR-RFLP of 16S-rRNA of each colony will be discussed.
